LDB1 is a widely expressed nuclear protein that participates in the formation of large multi-component complexes with important functions in cell-fate determination, cytoskeletal organization, and tissue development. The encoded protein also participates in chromatin looping and transcription activation. In primary erythroid cells, LDB1 helps facilitate long-range enhancer-promoter interactions in the β-globin locus and forms a core component of transcription complexes (such as TAL1, LMO2 and GATA1) that physically link the β-globin LCR (LCRB) and β-like globin genes during erythropoiesis. LDB1 complexes have a global role in erythroid gene transcription, and have been shown to bind to genes involved in a wide range of erythroid cellular processes including transcription (eg, Klf1, Zfpm1/Fog1, Nfe2), heme synthesis (eg, Alad, Alas2, Cpox, Fech, Ppox), and cytoskeletal organization (eg, Add1/2, Ank1, Epb4.1, Epb4.2, Spna1). LDB1 complexes bind and activate genes that are induced by GATA1, indicating that GATA1 functions in the context of an LDB1 complex to activate transcription of target genes. LDB1 complexes are involved in the activation of KLF1 through binding to a conserved E-box/GATA motif within the KLF1 promoter. LDB1 complex binding sites frequently colocalize with KLF1 binding sites and with consensus binding motifs for other erythroid transcription factors.
