Description:
CBFA2T3 (commonly known as ETO2) is a transcriptional repressor that interacts with several members of histone deacetylases (HDACs) critical to facilitate transcriptional repression. It is part of a TAL-1-containing complex in erythroid cells, recruited through interaction with E2A/HEB. Other TAL-1 partners include GATA-1, LMO2 and Ldb1. This complex occupies the β-globin locus control region (βLCR) and the BGL3 non-coding transcript region downstream of the HBG1 gene. Transcription of the BGLT3 locus is critical for γ-globin gene long-range looping interactions with the β-LCR. The γ-globin/BGL3 region is also occupied by BCL11A, a well-established negative regulator of fetal haemoglobin (HbF) expression. Knock-down of ETO2 upregulates γ-globin and BGL3 transcription in erythroid K562 cells. Ectopic expression and siRNA knockdown experiments in haematopoietic progenitor cells show that ETO2 actively represses erythroid TAL-1 target genes and regulates the expansion of erythroid progenitors.
