Description:
MicroRNAs (miRNAs), such as MIR15A, are small, non-coding RNAs that play key roles in the post-transcriptional regulation of gene expression by targeting specific mRNAs. MIR15A is located on chromosome 13q14.3 within DLEU2 gene, adjacent to microRNA MIR16-1 [GeneID: 459]; both microRNAs have identical seed sequences. MIR15A/MIR16-1 are strongly expressed in primary adult erythroid cells and contribute to the process of erythroid differentiation. Their expression correlates with increased embryonic and foetal haemoglobin levels in newborns in human trisomy 13 through direct inhibition of MYB [GeneID: 43], a potent negative regulator of foetal haemoglobin (HbF) expression. MIR15A expression correlates with increased HbF synthesis in erythroid cells of beta-thalassaemia patients. Therapeutic approaches that increase the HbF concentration have great potential for ameliorating the clinical and haematologic severity of beta-thalassaemia and other beta-haemoglobinopathies.
