Description:
MicroRNAs (miRNAs), such as MIR122, are small, non-coding RNAs that play key roles in the post-transcriptional regulation of gene expression by targeting specific mRNAs. MIR122 comprises nearly 70% of all hepatic miRNA expression and plays a key role in the regulation of systemic iron metabolism primarily through an effect on hepcidin levels. In fact, it directly targets and suppresses the hemochromatosis (HFE) and hemojuvelin (HJV or HFE2) genes that encode activators of hepcidin expression. Hepcidin deficiency results in the development of systemic iron overload because of excessive iron absorption. Blood transfusion was shown to affect MIR122 expression, resulting in higher levels of MIR122 in transfusion-dependent (TDT) β-thalassemia patients that were also associated with excess iron in liver and cardiac tissues. It is postulated that MIR122 could be a biomarker for cellular damage under condition of tissue iron excess.
