Description:
Guanine nucleotide-binding proteins (G proteins; stimulatory (Gs) or inhibitory (Gi)) are heterotrimeric proteins composed of α-, β-, and γ-subunits. The encoded protein of the GNAI2 gene is an alpha subunit of Gi proteins, which are implicated in the inhibition of adenylate cyclase, the enzyme that generates the second messenger cyclic AMP. GNAI2 is implicated in the regulation of the MAP kinase signaling pathway, which plays a key role in the regulation of erythropoiesis. Ineffective erythropoiesis due to intramedullary apoptosis and delayed maturation of erythroid progenitor cells is the hallmark of beta-thalassaemia. A significant change in the expression of the GNAI2 gene has been reported in patients with beta-thalassaemia, thus implicating GNAI2 in the pathogenesis of beta-thalassaemia, likely through the MAP kinase pathway.
