Description:
The protein encoded by this gene belongs to a family of DNA-binding proteins that includes the AP-1 family of transcription factors, cAMP-response element binding proteins (CREBs) and CREB-like proteins. These proteins bind DNA via their basic region and dimerize via their leucine zipper domain to form a variety of homo- and heterodimers to regulate gene transcription. CRISPR-Cas screening in HUDEP-2 cells identified ATF4 as a regulator of γ-globin transcription through direct transcriptional control of BCL11A, a known repressor of γ-globin transcription. ATF4 stimulates transcription of BCL11A by binding to its enhancer (+55) and increasing enhancer-promoter contacts. ATF4 is regulated by HRI [GeneID: 264], and serves as a critical component in γ-globin silencing (linear signaling pathway from HRI to ATF4 to BCL11A to γ-globin).
