THPO gene encodes a haematopoietic factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. It is synthesized as a 332 amino acid glycoprotein in the liver. It binds to receptors that dimerize and activate cytoplasmic kinases, which in turn phosphorylate downstream signalling molecules. It may be the major physiological regulator of circulating platelets. Genetic variance in THPO gene associated with haematological parameters (e.g., platelet count).
