This gene encodes an RNA binding protein that recruits target transcripts to cytoplasmic protein-RNA complexes (mRNPs). It also modulates the rate and location at which target transcripts encounter the translational apparatus and shields them from endonuclease attacks or microRNA-mediated degradation. IGF2BP1 was demonstrated to regulate fetal-to adult developmental transition in human erythroblasts. IGF2BP1 is a target of the lethal-7 (let-7) miRNA family, such that IGF2BP1 is expressed during early development and steadily decreases after birth in most tissues. Overexpression of IGF2BP1 in cultured human adult erythroblasts from beta-thalassaemia and healthy donors resulted in elevated HbF levels. Similarly, IGF2BP1 suppressed βS-globin and increased γ-globin expression in SCD patient-derived erythroblasts. IGF2BP1 promotes Interaction between the βLCR andγ-globin genes. IGF2BP1 is a regulator of BCL11A and HMGA2, which are both implicated in the modulation of Hb F expression.
