Description:
The protein encoded by this gene belongs to a distinct class of adenylyl cyclases (ACs) that is soluble and insensitive to G protein or forskolin regulation. Soluble AC (sAC) is encoded by a single functional sAC gene, the ADCY10. ACs catalyse the conversion of ATP to the second messenger cyclic AMP (cAMP) that mediates numerous biological responses, including transcriptional regulation, metabolism and cell migration. In contrast to membrane-associated AC, sAC is thought to generate cAMP closer to its required site of action. sAC activity requires divalent metal cations in the catalytic active site of the enzyme in order to coordinate the binding and cyclizing of ATP and it is also uniquely activated through bicarbonate binding. sAC is implicated in the regulation of cell growth, particularly in hyperplasia, and plays a central role in sperm physiology. sAC contributes to the regulation of CREB-mediated Na+/K+-ATPase expression in the vascular endothelium and is an important regulator of endothelial stiffness. A variation in ADCY10 associated with Hb F response to hydroxyurea in sickle cell disease cohorts.
