Description:
The ITGA11 gene encodes an integrin α-subunit. Integrins are transmembrane receptors composed of one α- and one β-subunit. The integrin family consists of 18 α- and 8 β-subunits that can assemble into 24 different heterodimers of distinct ligand-binding specificity and tissue distribution. The encoded protein contains an I domain (also called the A domain), which is central in ligand binding. ITGA11 facilitates the movement of cells through a collagen-rich extracellular matrix. It is expressed mainly in regions of forming cartilage, bone, invertebral discs and cornea during development, and dimerizes with the β1-subunit to form the collagen receptor α11β1. It binds type I and type IV collagen. Collagen receptors (α1β1, α2β1, α10β1, and α11β1) are involved in various biological processes, including mesenchymal stem cell proliferation, wound healing, and innate and acquired immunity. ITGA11 plays a role in myofibroblast differentiation with implications for fibrotic disease, and is regulated by the hedgehog signalling pathway. It is needed in the periodontal ligament to facilitate the movement of periodontal ligament fibroblasts and the reorganization of collagen during tooth eruption. It may be involved in attaching muscle tissue to the extracellular matrix. Alternative transcriptional splice variants have been found for this gene, but their biological validity is not determined. An ITGA11 variant associated with leukocytosis in sickle cell anaemia.
