Description:
The FOXO3 gene encodes a member of the class 'O' of forkhead-box (FOX) proteins (hence, FOXO). FOX proteins constitute a large family of transcription factors, which play a key role in the regulation of crucial biological processes, such as cellular proliferation, metabolism, inflammation, stress resistance, and apoptosis. FOX proteins are defined by a characteristic 'forkhead' box, a sequence of about 100 amino acids forming a motif that binds to DNA. This motif is also known as a 'winged-helix' DNA binding domain given its distinct butterfly-like appearance. FOXOs have four members and are tightly controlled by post-translational modifications. FOXO3 is expressed in nearly all tissues. FOXO3 protects cells against oxidative stress by increasing the production of ROS scavenging enymes, such as MnSOD and catalase. It has been shown to protect erythropoiesis against oxidative stress. Resveratrol, a natural phytoalexin with antioxidant properties used in β-thalassemia treatment, has been shown to activate FOXO3. FOXO3 is also critical for hematopoietic self-renewal. Translocation of this gene with the MLL gene is associated with secondary acute leukemia. Alternatively spliced transcript variants encoding the same protein have been observed.
