NFIX belongs to the nuclear factor I (NFI) family of transcription factors and binds the palindromic sequence 5'-TTGGCNNNNNGCCAA-3' present in viral and cellular promoters. NFI proteins consist of a conserved N-terminal DNA-binding and dimerization domain and a C-terminal transactivation/repression domain. They exhibit a high variability due to extensive alternative splicing. NFIX is highly expressed by haematopoietic stem and progenitor cells (HSPCs) and plays an important role in regulating their repopulating potential and survival. It has been shown to regulate myeloid-vs-lymphoid cell fate at early stages of HSPC development, potentially via the direct modulation of lineage-specific transcription factors.
